
The Caco-2 permeability assay is a widely used in vitro method to assess the permeability of drugs and other compounds across the intestinal epithelium. Caco-2 cells are derived from a human colon carcinoma cell line and possess many characteristics of mature enterocytes, making them a suitable model for studying drug absorption in the intestine.
In the assay, Caco-2 cells are grown on a permeable membrane, forming a monolayer that mimics the intestinal epithelium. The compound of interest is then applied to the apical (luminal) side of the monolayer, and the amount that crosses the monolayer and appears in the basolateral (blood) side is measured.
The permeability of the compound is determined by calculating the apparent permeability coefficient (Papp), which takes into account the concentration of the compound on both sides of the monolayer, the surface area of the monolayer, and the time of incubation. Papp values can be used to classify compounds into different categories based on their absorption potential, such as high, moderate, or low permeability.
